Linkage relationships between the Mj-1 locus and AFLP and AFLP-derived markers were estimated using MAPMAKER version 3.0 for IBM-PC (Lander et al., 1987; Lincoln et al. 1992). Linkage groups were first assigned based upon two-point analysis via ‘group’ command. Markers were initially associated with two-point comparisons of the group command (threshold LOD score = 5.0 and recombination fraction 0.25). The most likely order of the markers was obtained using exhaustive multipoint analysis via ‘compare’ command and the final maximum likelihood map was obtained with ‘map’ command. Recombination frequencies were transformed to Kosambi map distances (Kosambi, 1944).
Figure 6.2. The AFLP linkage map of the Mj-1 (Meloidogyne javanica resistance) locus in carrot. The map population was composed by 188 F2 individuals from the cross ‘Brasilia-1252’ (resistant parent) × ‘6274B’ (susceptible parent). The genetic distance in cM (Kosambi function) between pairs of markers is shown at the left and the AFLP marker designation is shown on the right
